A loop-mediated isothermal amplification assay for the rapid diagnosis of soybean rust caused by Phakopsora pachyrhizi

Abstract Soybean rust caused by the fungus Phakopsora pachyrhizi is one of most destructive diseases to soybean production worldwide. For rapid diagnosis and control rust, we developed a loop-mediated isothermal amplification (LAMP) assay that targets P . -specific gene ( Phapa_6409908 ) identified from comparative genomic analysis 61 Pucciniomycotina strains. Using set screened primers optimized reaction conditions 62°C for 70 min, LAMP can detect while excluding other plant pathogens. The consistently showed high sensitivity in detecting low contents DNA (10 pg). We confirmed practical application this diagnosing using leaf samples collected fields four cities Sichuan Province, China, 2020 2021. provides specific, sensitive, method , which assist prediction, prevention, major disease.
